正己醛对黄芪不定根生长及次生代谢产物积累的影响

摘 要:

本文以黄芪不定根为实验材料, 通过在培养基中添加正己醛的方式, 以生物量、多糖和总皂苷含量为评价指标, 研究了正己醛对黄芪生长和次生代谢的影响。结果表明: (1)正己醛显著影响黄芪不定根固体培养物的生长, 10 μmol•L-1正己醛具抑制作用, 而50 μmol•L-1有促进作用; (2)除添加50 μmol•L-1的处理外, 黄芪不定根液体培养物中的多糖含量普遍高于固体培养物中的; (3) 50 μmol•L-1正己醛对多糖合成的影响与不定根培养方法有关, 在液体悬浮培养体系中具抑制作用, 但在固体培养体系中起促进作用; (4)固体培养物中皂苷含量普遍较高且正己醛的影响较小, 而液体培养体系中50 μmol•L-1正己醛能促进黄芪不定根合成皂苷类物质。由此可见, 正己醛对黄芪不定根生长和次生代谢的影响不仅与正己醛浓度有关, 还与不定根培养方法有关, 固体培养体系中添加50 μmol•L-1正己醛对于多糖和皂苷类物质的合成效果最佳。

Effects of Hexanal on Growth and Accumulation of Secondary Metabolites in Astragalus membranaceus var. mongholicus Adventitious Roots

Abstract:

In this study, effects of hexanal on growth and secondary metabolism of Astragalus membranaceus var. mongholicus adventitious roots was investigated by adding it into the medium before culture. The culture had been continued for 4 weeks. After that, biomass and contents of polysaccharide and total saponins in the cultures were determined and used to evaluate its effects. The results showed that hexanal obviously affected the growth of the solid cultures. Specifically, 10 μmol•L-1 of hexanal resulted in a significant decrease in the biomass of the solid cultures while 50 μmol•L-1 of hexanal led to a significant increase. With respect to the polysaccharide content, its value in the liquid cultures was generally higher than that in the solid cultures excluding these supplied with 50 μmol•L-1 of hexanal. It is of interest that the supplement of 50 μmol•L-1 of hexanal in liquid medium exerted a negative role on the accumulation of polysaccharide but in solid medium a positive one. Although no significant difference in the content of total saponins in the solid cultures was observed between the control and the supplement of hexanal, their levels were generally higher than that in the liquid cultures. Meanwhile, supplemental 50 μmol•L-1 of hexanal in liquid medium resulted in a significant increase in the level of total saponins. Altogether, hexanal was involved in the growth and accumulation of secondary metabolites of A. membranaceus var. mongholicus adventitious roots in a dose- and culture method-dependent way, and adding 50 μmol•L-1 of hexanal into the solid medium is preferred for the production of both polysaccharide and total saponins.
